Extensive admixture in Brazilian sickle cell patients: implications for the mapping of genetic modifiers.

چکیده
This study was developed after approval by the Ethical Committees from the Universidade Federal de Minas Gerais and the Hemominas Foundation. We estimated admixture in unrelated subjects with sickle cell disease (SCD, n = 200) and healthy blood donors (n = 291), sampled across different regions of the state of Minas Gerais (Figure S1), a geographic area in South Eastern Brazil similar to that of France, peopled by more than 20 million inhabitants. The Minas Gerais State had a very important role during the colonization of Brazilian inlands, and several cities were funded during the XVIII Century, when due to an intensive mining activity, this state was the economical center of Brazil.1 The slaves that arrived to Minas Gerais had a diversified African origin: those coming from Rio de Janeiro were mainly from Angola, and those coming from Bahia were mainly from Ivory Coast.2 Admixture studies on healthy individuals from specific locations of Minas Gerais suggest extensive admixture.3,4,5

Level of Hemoglobin F and Gg Gene Expression in Sickle Cell Disease and Their Association with Haplotype and XmnI Polymorphic Site in South of Iran
Background: Molecular genetic factors regulating hemoglobin F (Hb F) expression are important modifiers of the severity of sickle cell anemia (SS). Methods: The prevalence of XmnI polymorphic site, the Gg:Ag ratio and the Hb F level were determined using PCR-RFLP procedure, HPLC and alkaline denaturation method, respectively, in various haplotypes of 52 patients with SS, 18 patients with sickle...
متن کاملElucidation of βs/ Globin Gene clusters Haplotypes Related to Sickle Cell Anemia in Khuzestan Province, Southwest of Iran
Background & objectives: The researcher clarified that β/Globin gene cluster haplotypes in patients with sickle cell anemia provide useful population data as predictors of the disease severity, gene flow, and the origins of sickle cell mutation in this region. Materials and methods: A total of 150 subjects was investigated in two different groups for five polymorphism restriction site...
متن کاملSteady state hemoglobin concentration and packed cell volume in homozygous sickle cell disease patients in Lagos, Nigeria
Background: Sickle cell disease is a genetic disorder of hemoglobin causing myriad of pathology including anemia. The purpose of this study was to evaluate the baseline values of steady state hemoglobin and packed cell volume as a guide to managing the early recognition of hemolytic crises in sickle cell anemia. Methods: A cross-sectional study was conducted among the sickle cell patients atte...
متن کاملGenetic modifiers of sickle cell disease.
Sickle cell disease is one of the best characterized human monogenic disorders. Complex genotype/phenotype correlations clearly demonstrate the interaction of multiple genetic and environmental factors. In the last 20 years, scientific research has applied genetic approaches to dissect some of these modifiers. This review highlights the more recent genetic association studies that have been app...
متن کاملAntioxidant Enzymes and Acute Phase Proteins Correlate with Marker of Lipid Peroxide in Adult Nigerian Sickle Cell Disease Patients
Objective(s) Sickle cell disease is a genetic disorder characterized by chronic haemolytic anaemia. Haemoglobin S containing red blood cells may be susceptible to oxidative stress due to imbalance between production of reactive oxygen species and the countering effect of the various antioxidants present in the body. Materials and Methods We evaluated some antioxidant enzymes which include gl...
